RESUMO

Cilia are biological structures essential to drive the mobility of secretions and maintain the proper function of the respiratory airways. However, this motile self-cleaning process is significantly compromised in the presence of silicone tracheal prosthesis, leading to biofilm growth and impeding effective treatment. To address this challenge and enhance the performance of these devices, we propose the fabrication of magnetic silicone cilia, with the prospect of their integration onto silicone prostheses. The present study presents a fabrication method based on magnetic self-assembly and assesses the interaction behavior of the cilia array with biological mucus. This protocol allows for the customization of cilia dimensions across a wide range of aspect ratios (from 6 to 85) and array densities (from 10 to 80 cilia/mm2) by adjusting the fabrication parameters, offering flexibility for adjustments according to their required characteristics. Furthermore, we evaluated the suitability of different cilia arrays for biomedical applications by analyzing their interaction with bullfrog mucus, simulating the airways environment. Our findings demonstrate that the fabricated cilia are mechanically resistant to the viscous fluid and still exhibit controlled movement under the influence of an external moving magnet. A correlation between cilia dimensions and mucus wettability profile suggests a potential role in facilitating mucus depuration, paving the way for further advancements aimed at enhancing the performance of silicone prostheses in clinical settings.

OBJECTIVES: Compared to lung resections, airway procedures are relatively rare in thoracic surgery. Despite this, a growing number of dedicated airway centres have formed throughout Europe. These centres are characterized by a close interdisciplinary collaboration and they often act as supra-regional referring centres. To date, most evidence of airway surgery comes from retrospective, single-centre analysis as there is a lack of large-scale, multi-institutional databases. METHODS: In 2018, an initiative was formed, which aimed to create an airway database within the framework of the ESTS database (ESTS-AIR). Five dedicated airway centres were asked to test the database in a pilot phase. A 1st descriptive analysis of ESTS-AIR was performed. RESULTS: A total of 415 cases were included in the analysis. For adults, the most common indication for airway surgery was post-tracheostomy stenosis and idiopathic subglottic stenosis; in children, most resections/reconstructions had to be performed for post-intubation stenosis. Malignant indications required significantly longer resections [36.0 (21.4-50.6) mm] when compared to benign indications [26.6 (9.4-43.8) mm]. Length of hospital stay was 11.0 (4.1-17.3) days (adults) and 13.4 (7.6-19.6) days (children). Overall, the rates of complications were low with wound infections being reported as the most common morbidity. CONCLUSIONS: This evaluation of the 1st cases in the ESTS-AIR database allowed a large-scale analysis of the practice of airway surgery in dedicated European airway centres. It provides proof for the functionality of ESTS-AIR and sets the basis for rolling out the AIR subsection to all centres participating in the ESTS database.

Suicide notes are an important warning sign for suicidal behaviors. In the first, a quantitative investigation, the statistical relationship of writing a suicide note: To the occurrence of lifetime suicide attempts, to lifetime self-harming behaviors, and to suicidal ideation in the two weeks prior to the assessment was evaluated. In the second study, a qualitative investigation, the content recalled by individuals regarding a suicide note was analyzed. In the quantitative study, 841 adults aged between 18 and 65 years, and 1,012 young adults participated. In the qualitative study, 18 young adults participated. Findings of the quantitative study reveal that writing a suicide note significantly related to the lifetime presence of a suicide attempt and to self-harming behaviors and to suicidal ideation in the two weeks prior to the assessment. Findings of the qualitative study suggest that individuals who have written suicide notes exhibit a significant self-oriented focus, yet they also demonstrate a strong sense of concern for the survivors. The surge in global polymeric waste underscores the imperative for biodegradable materials to substitute traditional polymers. Crucially, advancements are needed for emerging technologies like Materials Extrusion (ME) in additive manufacturing, where current biodegradable materials exhibit limitations. This work was based on the development of a biodegradable composite filament. The inner of the mango seed (kernel) and poly(3-hydroxybutyrate-co-3-hydroxyvalerate) (PHBV) were used as raw materials. The properties of PHBV and mango by-product mixture were first evaluated by direct-extrusion printing. Then, the feasibility of manufacturing the filaments was studied. Initially, the kernel seed mango was characterized thermally, chemically, and morphologically by DSC, FTIR, and SEM, respectively. It was observed that the addition of mango by-product contributed to the decrease of PHBV crystallinity, resulting in the reduction of printed parts retraction and increases the Tg, as shown by the DMA. The structure of the native starch was preserved due to non-gelatinization, even after processing steps, as indicated by thermal, chemical, and morphological analyses. Finally, PHBV filaments containing mango by-products were fabricated, and prototypes were manufactured by ME to demonstrate the potential for market acceptance and commercialization of the studied filament.

ABSTRACT Despite the excellent properties of silicone endotracheal prostheses, their main limitation is the formation of a polymicrobial biofilm on their surfaces. It can cause local inflammation, interfering with the local healing process and leading to further complications in the clinical scenario. The present study evaluated the inhibitory effect of cold atmospheric plasma (CAP) on multispecies biofilms grown on the silicone protheses' surfaces. In addition to silicone characterization before and after CAP exposure, CAP cytotoxicity on immortalized human bronchial epithelium cell line (BEAS-2B) was evaluated. The aging time test reported that CAP could temporarily change the silicone surface wetting characteristics from hydrophilic (80.5°) to highly hydrophilic (<5°). ATR-FTIR showed no significant alterations in the silicone surficial chemical composition after CAP exposure for 5 min. A significant log reduction in viable cells in monospecies biofilms (log CFU/mL) of C. albicans, S. aureus, and P. aeruginosa (0.636, 0.738, and 1.445, respectively) was detected after CAP exposure. Multispecies biofilms exposed to CAP showed significant viability reduction for C. albicans and S. aureus (1.385 and 0.831, respectively). The protocol was not cytotoxic to BEAS-2B. CAP can be a simple and effective method to delay multispecies biofilm formation inside the endotracheal prosthesis.

OBJECTIVES: The Brazilian Society of Thoracic Surgeons conducted an online survey to determine the number of surgeons that perform adult and paediatric airway surgery and to understand the practice patterns along the country. METHODS: Active members were electronically invited to complete the questionnaire through the REDCapR platform. Invitations were sent from January to April 2020. The survey encompassed 40 questions that explored 4 different topics in the assessment of tracheal diseases: (i) surgeon's demography; (ii) institutional profile, (iii) education and training in laryngo-tracheal surgery and (iv) preoperative and postoperative evaluation. RESULTS: Eighty-nine percentage of the responders declared to perform tracheal surgery with a median of 5 tracheal resection procedures per year [interquartile range (IQR) 3-12]. Interaction with other specialties occurs in 37.3% of cases. Access to technology and devices is highly variable across the country. Resident training in airway surgery consists in traditional lectures in 97% of the cases. Training in animals (15.2%), cadavers (12.1%) and simulators (6.1%) are rare. Preoperatory evaluation encompasses flexible bronchoscopy (97.8%) and/or computed tomography (CT) scan of the airways (90.6%). Swallowing (20.1%) and voice (14.4%) disorders are rarely evaluated. Eighty-nine percentage of the surgeons consider bronchoscopy to be the preoperatory gold-standard exam, followed by CT scan (38.8%) and CT-3D reconstruction (37.4%). CONCLUSIONS: Brazilian surgeons refer that airway resection and reconstruction are part of their current practice, but the total number of procedures per surgeon per year is low. Access to high-end technology and equipment is heterogenous. Training offered to residents in most academic institutions relies on traditional lectures.

Background: Central airway diseases requiring frequent outpatient visits to a specialized medical center due to tracheal devices. Many of these patients have mobility and cognition restrictions or require specialized transport due to the need for supplemental oxygen. This study describes the implementation and results of a telemedicine program dedicated to patients with central airway diseases based in a Brazilian public health system. Methods: A retrospective study of telemedicine consultation for patients with central airway diseases referred to a public academic hospital between August 1, 2020 and August 1, 2022. The consultations occurred in a telemedicine department using the hospital's proprietary platform. Data retrieved consisted of demographics, disease characteristics, and the treatment modalities of the patients. The analysis included the savings in kilometers not traveled, the carbon footprint based on reducing CO2 emissions, and the cost savings in transportation. Results: A total of 1,153 telemedicine visits conducted in 516 patients (median age of 31.5 years). Two hundred ninety patients (56.2%) had a tracheal device (129 silicone T-Tube, 128 tracheostomy, and 33 endoprosthesis) and 159 patients (30.8%) had difficulties in transportation to the specialized medical center. Patients were served from 147 Brazilian cities from 22 states. The savings in kilometers traveled was 1,224,108.54 km, corresponding to a 250.14 ton reduction in CO2 emissions. The costs savings in transportation for the municipalities was BRL$ 1,272,283.78. Conclusions: Telemedicine consultations for patients with central airway diseases are feasible and safe. Cost savings and the possibility of disseminating specialized care make telemedicine a fundamental tool in current medical practice.

In the context of Shared Autonomous Vehicles, the need to monitor the environment inside the car will be crucial. This article focuses on the application of deep learning algorithms to present a fusion monitoring solution which was three different algorithms: a violent action detection system, which recognizes violent behaviors between passengers, a violent object detection system, and a lost items detection system. Public datasets were used for object detection algorithms (COCO and TAO) to train state-of-the-art algorithms such as YOLOv5. For violent action detection, the MoLa InCar dataset was used to train on state-of-the-art algorithms such as I3D, R(2+1)D, SlowFast, TSN, and TSM. Finally, an embedded automotive solution was used to demonstrate that both methods are running in real-time.

Tracheal replacement with a bioengineered tracheal substitute has been developed for long-segment tracheal diseases. The decellularized tracheal scaffold is an alternative for cell seeding. It is not defined if the storage scaffold produces changes in the scaffold's biomechanical properties. We tested three protocols for porcine tracheal scaffold preservation immersed in PBS and alcohol 70%, in the fridge and under cryopreservation. Ninety-six porcine tracheas (12 in natura, 84 decellularized) were divided into three groups (PBS, alcohol, and cryopreservation). Twelve tracheas were analyzed after three and six months. The assessment included residual DNA, cytotoxicity, collagen contents, and mechanical properties. Decellularization increased the maximum load and stress in the longitudinal axis and decreased the maximum load in the transverse axis. The decellularization of the porcine trachea produced structurally viable scaffolds, with a preserved collagen matrix suitable for further bioengineering. Despite the cyclic washings, the scaffolds remained cytotoxic. The comparison of the storage protocols (PBS at 4 °C, alcohol at 4 °C, and slow cooling cryopreservation with cryoprotectants) showed no significant differences in the amount of collagen and in the biomechanical properties of the scaffolds. Storage in PBS solution at 4 °C for six months did not change the scaffold mechanics.

Biological nitrogen fixation in soybean is enhanced when seed is treated with cobalt (Co) and molybdenum (Mo) prior to planting. In this study, our objective was to verify if Co and Mo application during the reproductive phase of the crop increases seed Co and Mo concentration without adverse effects on seed quality. Two experiments were conducted. First, we investigated foliar and soil Co and Mo application under greenhouse conditions. Next, we validated the results obtained in the first study. The treatments for both experiments consisted of Co doses combined with Mo, and a control without Co and Mo application. The foliar application was more efficient in producing enriched Co and Mo seed; meanwhile, as Co doses increased so did Co and Mo concentrations in the seed. There were no adverse effects on nutrition, development, quality, and yield of parent plants and seed when these micronutrients were applied. The seed showed higher germination, vigor, and uniformity for the development of soybean seedlings. We concluded that the application of 20 g ha-1 Co and 800 g ha-1 Mo via foliar application at the reproductive stage of soybean increased germination rate and achieved the best growth and vigor index of enriched seed.

Salinity constitutes a major abiotic factor that negatively affects crop productivity. Inoculation with plant growth-promoting bacteria (PGPB) is proven to increase plant tolerance to abiotic stresses and enhance plant growth, development and productivity. The present study aims to increase the resilience of crops to salinity using bacteria from the microbiome of plants growing in saline environments. For that, the halotolerance of bacteria present in the roots of natural plants growing on Sal Island, which is characterized by its arid environment and maritime influence, was determined, with some strains having extreme halotolerance. Their ability to produce plant growth-promoting traits was evaluated, with most strains increasing indole acetic acid (26-418%), siderophore (>300%) and alginate (2-66%) production and phosphate solubilization (13-100%) under salt stress. The strains evidencing the best performance were inoculated in maize (Zea mays L.) plants and their influence on plant growth and biochemical status was evaluated. Results evidenced bacterial ability to especially increase proline (55-191%), whose osmotic, antioxidant and protein-protecting properties reduced protein damage in salt-stressed maize plants, evidencing the potential of PGPB to reduce the impact of salinity on crops. Enhanced nutrition, phytohormone production and osmolyte synthesis along with antioxidant response all contribute to increasing plant tolerance to salt stress.

The impact of hazardous materials, such as Hg, on life is far from being understood and due to the high number of polluted sites it has generated great concern. A biochemical and lipidomic approach was used to assess the effects of Hg on the saltmarsh halophyte Halimione portulacoides. Plants were collected at two sites of a Hg contaminated saltmarsh. Hg accumulation and distribution in the plant, biochemical parameters (antioxidant and metabolic) and lipid profiles were determined and compared between plant organs and sites (s1 and s2). Hg did not induce antioxidant enzyme activity. Lipid profiles changed under Hg exposure, especially in leaves, decreasing the unsaturation level, the membrane fluidity and stability, and evidencing that membrane lipid remodeling influences plant tolerance to Hg. This knowledge can help select the most appropriate methodologies for the restoration of Hg polluted hotspots, curtailing a serious environmental problem threatening saltmarshes.

Contamination with Arsenic, a toxic metalloid, is increasing in the marine environment. Additionally, global warming can alter metalloids toxicity. Polychaetes are key species in marine environments. By mobilizing sediments, they play vital roles in nutrient and element (including contaminants) cycles. Most studies with marine invertebrates focus on the effects of metalloids on either adults or larvae. Here, we bring information on the effects of temperature increase and arsenic contamination on the polychaete Hediste diversicolor in different growth stages and water temperatures. Feeding activity and biochemical responses-cholinesterase activity, indicators of cell damage, antioxidant and biotransformation enzymes and metabolic capacity-were evaluated. Temperature rise combined with As imposed alterations on feeding activity and biochemical endpoints at different growth stages. Small organisms have their antioxidant enzymes increased, avoiding lipid damage. However, larger organisms are the most affected class due to the inhibition of superoxide dismutase, which results in protein damage. Oxidative damage was observed on smaller and larger organisms exposed to As and temperature of 21 °C, demonstrating higher sensibility to the combination of temperature rise and As. The observed alterations may have ecological consequences, affecting the cycle of nutrients, sediment oxygenation and the food chain that depends on the bioturbation of this polychaete.

Climate change and anthropogenic activities are responsible for extensive crop yield losses, with negative impact on global agricultural production. The occurrence of extreme weather events such as drought is a big challenge for agriculture, negatively impacting crops. Thus, methodologies reducing crop dependence on water will be a great advantage. Plant roots are colonized by soil bacteria, that can establish beneficial associations with plants, increasing crop productivity and plant tolerance to abiotic stresses. The aim of this study was to promote plant growth and to increase crop tolerance to drought by inoculation with osmotolerant bacterial strains. For that, bacteria were isolated from plants growing in Sal Island (Cape Verde) and identified. The osmotolerance and plant-growth promotion (PGP) abilities of the strains were determined. A maize seed cultivar tolerant to drought was inoculated with the strains evidencing best PGP capacity and osmo-tolerance. Results evidenced the ability of some bacterial strains increasing the development and inducing osmotolerance in plants. These results evidence the potential of osmotolerant bacteria to further increase the level of tolerance of maize varieties tolerant to drought, decreasing the dependence of this crop on irrigation, and open new perspectives to growth maize in drought affected areas and to use water more efficiently.

Plant roots are colonized by rhizobacteria, and these soil microorganisms can not only stimulate plant growth but also increase tolerance to stress through the production of volatile organic compounds. However, little is known about the effect that these plant beneficial volatiles may have on bacteria. In this study, the effects on growth and oxidative status of different concentrations of three volatiles already reported to have a positive influence on plant growth (2-butanone, 3-methyl-1-butanol, and 2,3-butanediol) were determined in A. thaliana and Rhizobium sp. strain E20-8 via airborne exposure in the presence and absence of Cd. It was expected to ascertain if the plant and the bacterium are influenced in the same way by the volatiles, and if exposure to stress (Cd) shifts the effects of volatiles on plants and bacteria. Results showed the antioxidant activity of the volatiles protecting the plant cell metabolism from Cd toxicity and increasing plant tolerance to Cd. Effects on bacteria were less positive. The two alcohols (3-methyl-1-butanol and 2,3-butanediol) increased Cd toxicity, and the ketone (2-butanone) was able to protect Rhizobium from Cd stress, constituting an alternative way to protect soil bacterial communities from stress. The application of 2-butanone thus emerges as an alternative way to increase crop production and crop resilience to stress in a more sustainable way, either directly or through the enhancement of PGPR activity.

RESUMO

With the evolution of technology associated with mobility and autonomy, Shared Autonomous Vehicles will be a reality. To ensure passenger safety, there is a need to create a monitoring system inside the vehicle capable of recognizing human actions. We introduce two datasets to train human action recognition inside the vehicle, focusing on violence detection. The InCar dataset tackles violent actions for in-car background which give us more realistic data. The InVicon dataset although doesn't have the realistic background as the InCar dataset can provide skeleton (3D body joints) data. This datasets were recorded with RGB, Depth, Thermal, Event-based, and Skeleton data. The resulting dataset contains 6 400 video samples and more than 3 million frames, collected from sixteen distinct subjects. The dataset contains 58 action classes, including violent and neutral (i.e., non-violent) activities.
